Nitric oxide (NO) side of lymphatic flow and immune surveillance.

چکیده

T he loss of cancer or bacteria detection by the immune system is traditionally looked at from the point of cell–cell or humoral mediator–cell interactions. However, immune surveillance in lymphatics critically requires cell transport along the lymphatics for entry of antigen/dendritic cells into the draining lymph node (1). Reductions in collecting lymphatic function affect all subsequent immune response. The work by Liao et al. (2), from a team wellversed in the complexities and limitations of transport in tumors, brings to light that lymphatic transport can be limited by a mechanism that is controlled by nitric oxide (NO) derived from bone marrow myeloid cells.

Confocal image-based computational modeling of nitric oxide transport in a rat mesenteric lymphatic vessel.

The lymphatic system plays important roles in protein and solute transport as well as in the immune system. Its functionality is vital to proper homeostasis and fluid balance. Lymph may be propelled by intrinsic (active) vessel pumping or passive compression from external tissue movement. With regard to the former, nitric oxide (NO) is known to play an important role modulating lymphatic vessel...

Nitric oxide permits hypoxia-induced lymphatic perfusion by controlling arterial-lymphatic conduits in zebrafish and glass catfish.

The blood and lymphatic vasculatures are structurally and functionally coupled in controlling tissue perfusion, extracellular interstitial fluids, and immune surveillance. Little is known, however, about the molecular mechanisms that underlie the regulation of bloodlymphatic vessel connections and lymphatic perfusion. Here we show in the adult zebrafish and glass catfish (Kryptopterus bicirrhis...

Effect of nitric oxide on skin blood flow of intact and morphine- dependent rats

Introduction: The relation between morphine and nitric oxide release has been shown. Due to important role of nitric oxide in regulation of skin blood flow, the aim of this study was to investigate the effect of nitric oxide synthase inhibitor (L-NAME) and nitric oxide synthesis precursor (L-arginine) on skin blood flow of intact and morphine-dependent rats. Methods: Skin blood flow of hind pa...

Role of Nitric Oxide and ATP-Sensitive K+ Channels in Regulation of Basal Blood Flow and Hypercapnic Vasodilatation of Cerebral Blood Vessels in Rabbit

Background: The mechanisms underlying cerebral hypercapnic vasodilatation are not fully understood. Objective: To investigate the role of nitric oxide (NO) and ATP-sensitive potassium (KATP) channels in basal blood flow regulation and hypercapnia-induced vasodilatation in rabbit cerebral blood vessels. Methods: The change in cerebral blood flow was measured by a laser Doppler flowmeter in 18 Ne...

Endothelial nitric oxide synthase regulates microlymphatic flow via collecting lymphatics.

Functional interactions between the initial and collecting lymphatics, as well as the molecular players involved, remain elusive. In this study, we assessed the influence of nitric oxide (NO) on lymphatic fluid velocity and flow, using a mouse tail model that permits intravital microscopy and microlymphangiography. We found that NO synthase (NOS) inhibition decreased lymphatic fluid velocity in...
